بهینه‌سازی بیان، خالص‌سازی و بررسی فعالیت آنزیم نوترکیب Tth DNA polymerase در E.coli BL21

کشف DNA پلی‌مرازهای مقاوم به حرارت از میکروارگانیسم‌های گرمادوست جهش بزرگی در مهندسی ژنتیک ایجاد کرد. آنزیم Tth DNA polymerase (Tth pol) از باکتری Thermus thermophiles جزو آنزیم‌های پرکاربرد در مهندسی ژنتیک است. خاصیت دوگانه پلیمرازی و رونوشت برداری معکوس، مقاومت به بازدارنده‌های PCR موجود در نمونه‌های زیستی، تکثیر سریع‌تر DNA در مقایسه با Taq DNA polymerase توجه محققان را به خود جلب کرده است. در این پژوهش، ژن کدکننده آنزیم Tth pol از باکتری T.thermophilus جدا شد، و با وکتور pTZ57R/T به باکتری E.coli DH5α منتقل شد. نتایج توالی‌یابی پلاسمیدهای نوترکیب صحت جداسازی ژن Tth pol را تأیید نمود. این ژن به وکتور بیانی pET28a الحاق شده و به میزبان بیانی E.coli BL21 منتقل شد. القای بیان با IPTG در غلظت‌های 8/0 و 1 میلی‌مولار، در دو محیط کشت حداقل تغییریافته و LB، و دمای 25 و 30 درجه سانتی‌گراد انجام شد. نمونه‌برداری تا 10 ساعت پس از القا و در فواصل زمانی 2 ساعته ادامه یافت. الکترفورز نمونه‌های پروتئینی بیشینه بیان را در دمای 30 درجه، غلظت 1 میلی‌مولار IPTG، محیط کشت حداقل تغییریافته و 10 ساعت پس از القا نشان داد. به دلیل وجود دنباله هیستیدینی در Tth pol نوترکیب از رزین نیکل برای خالص‌سازی استفاده شد. پس از خالص‌سازی فعالیت پلیمرازی Tth pol در PCR و فعالیت رونوشت برداری معکوس آن در سنتز cDNA بررسی شد. نتایج نشان داد این آنزیم دارای هر دو ویژگی می‌باشد. با توجه به نتایج، بهینه‌سازی شرایط کشت در مقیاس وسیع‌تر مانند فرمانتور به منظور بیان بیشتر Tth pol پیشنهاد می‌شود.
